Effect of energy supply on enzyme induction by pyrimidine requiring mutants of Escherichia coli.

چکیده

A study of the formation of induced (adaptive) enzymes in purine or pyrimidine requiring mutants of Escherichia coli was reported in a previous publication (Pardee, 1954). The idea that synthesis of ribonucleic acid (RNA) is essential for protein synthesis is supported by the inability of the bacterial mutants to form protein and several enzymes, including f3galactosidase, in the absence of purines or pyrimidines. All of these experiments were performed in a medium which contained an energy supply other than the inducer. Early in the work, it was noted, however, that in a medium which lacked both an energy supply and the nucleic acid component, formation of the induced enzyme did proceed to a considerable extent. The explanation of this phenomenon seems to lie in certain complications not pertinent to the relation of nucleic acid and protein synthesis and is the subject of this communication.

Study of Mutations in the DNA gyrase gyrA Gene of Escherichia coli

Quinolones are a large and widely consumed class of synthetic drugs. Expanded-spectrum quinolones, like ciprofloxacin are highly effective against Gram-negative bacteria, especially Escherichia coli. In E. coli the major target for quinolones is DNA gyrase. This enzyme is composed of two subunits, GyrA and GyrB encoding by gyrA and gyrB, respectively. Mutations in either of these genes cause qu...

The RihA, RihB, and RihC Ribonucleoside Hydrolases of Escherichia coli

Pyrimidine-requiring cdd mutants of Escherichia coli deficient in cytidine deaminase utilize cytidine as a pyrimidine source by an alternative pathway. This has been presumed to involve phosphorylation of cytidine to CMP by cytidine/uridine kinase and subsequent hydrolysis of CMP to cytosine and ribose 5-phosphate by a putative CMP hydrolase. Here we show that cytidine, in cdd strains, is conve...

Expression of tolC and organic solvent tolerance of Escherichia coli ciprofloxacin resistant mutants

AcrAB-TolC is a major efflux pump in Escherichia coli. It was reported that tolC is overexpressed and involves in improving the organic solvent tolerance level in Escherichia coli marR mutants that are resistant to several antibiotics, such as ciprofloxacin. Low and intermediate levels resistance did not improve organic solvent tolerance. Thus, in this descriptive-experimental study it was deci...
